    Bandwidth-efficient Video Streaming with Network Coding on Peer-to-Peer Networks

    PhDOver the last decade, live video streaming applications have gained great popularity among users but put great pressure on video servers and the Internet. In order to satisfy the growing demands for live video streaming, Peer-to-Peer(P2P) has been developed to relieve the video servers of bandwidth bottlenecks and computational load. Furthermore, Network Coding (NC) has been proposed and proved as a significant breakthrough in information theory and coding theory. According to previous research, NC not only brings substantial improvements regarding throughput and delay in data transmission, but also provides innovative solutions for multiple issues related to resource allocation, such as the coupon-collection problem, allocation and scheduling procedure. However, the complex NC-driven P2P streaming network poses substantial challenges to the packet scheduling algorithm. This thesis focuses on the packet scheduling algorithm for video multicast in NC-driven P2P streaming network. It determines how upload bandwidth resources of peer nodes are allocated in different transmission scenarios to achieve a better Quality of Service(QoS). First, an optimized rate allocation algorithm is proposed for scalable video transmission (SVT) in the NC-based lossy streaming network. This algorithm is developed to achieve the tradeoffs between average video distortion and average bandwidth redundancy in each generation. It determines how senders allocate their upload bandwidth to different classes in scalable data so that the sum of the distortion and the weighted redundancy ratio can be minimized. Second, in the NC-based non-scalable video transmission system, the bandwidth ineffi- ciency which is caused by the asynchronization communication among peers is reduced. First, a scalable compensation model and an adaptive push algorithm are proposed to reduce the unrecoverable transmission caused by network loss and insufficient bandwidth resources. Then a centralized packet scheduling algorithm is proposed to reduce the unin- formative transmission caused by the asynchronized communication among sender nodes. Subsequently, we further propose a distributed packet scheduling algorithm, which adds a critical scalability property to the packet scheduling model. Third, the bandwidth resource scheduling for SVT is further studied. A novel multiple- generation scheduling algorithm is proposed to determine the quality classes that the receiver node can subscribe to so that the overall perceived video quality can be maxi- mized. A single generation scheduling algorithm for SVT is also proposed to provide a faster and easier solution to the video quality maximization function. Thorough theoretical analysis is conducted in the development of all proposed algorithms, and their performance is evaluated via comprehensive simulations. We have demon- strated, by adjusting the conventional transmission model and involving new packet scheduling models, the overall QoS and bandwidth efficiency are dramatically improved. In non-scalable video streaming system, the maximum video quality gain can be around 5dB compared with the random push method, and the overall uninformative transmiss- sion ratio are reduced to 1% - 2%. In scalable video streaming system, the maximum video quality gain can be around 7dB, and the overall uninformative transmission ratio are reduced to 2% - 3%

    Association of lipid lowering drugs and the risk of systemic lupus erythematosus: a drug target Mendelian randomization

    Background and objective: An interaction between low-density lipoprotein level, lipid-lowering drugs, and systemic lupus erythematosus (SLE) was reported by previous studies. However, whether lipid-lowering drugs provided protective effect for reducing the risk of SLE was unclear. We aimed to clarify this causal relationship through a drug-target Mendelian randomization (MR) study.Methods: Genetic instruments—single nucleotide polymorphism (SNPs)—were utilized to proxy inhibition of the three genes—3-hydroxy-3-methylglutaryl-CoA reductase (HMGCR), proprotein convertase subtilisin/kexin type 9 (PCSK9), and Niemann-Pick C1-Like 1(NPC1L1), which was corresponded to three lipid-lowering drugs—statins, evolocumab, and ezetimibe. Low-density lipoprotein (LDL) cholesterol was selected as the biomarker for the measurement of the inhibitors of HMGCR, PCSK9, and NPC1L1, and the genetic data were acquired from the Global Lipids Genetics Consortium, which consisted of 1.3 million participants of European ancestry and 146.5 thousand participants of East Asian ancestry. The genetic dataset of SLE was acquired from two large-scale GWAS studies; one recruited 23,210 participants (7,219 SLE cases and 15,991 controls) of European ancestry and the other one recruited 12,653 participants (4,222 SLE cases and 8,431 controls) of Chinese ancestry. The primary analysis used the inverse variance weighted (IVW) method. Four additional sensitivity analyses, colocalization analysis, and stratification analysis were performed.Results: The primary analysis showed that inhibition of PCSK9 (evolocumab) was associated with a significantly lower risk of SLE [odds ratio (OR) 0.51, 95%CI 0.34 to 0.76, p = 0.001] in the European population. The secondary analyses had similar findings. Stratification analysis demonstrated that the preventive effect of PCSK9 inhibition for SLE was similar in both males and females. However, the results were not replicated in the East Asian population. The inhibition of HMGCR (statins) and NPC1L1 (ezetimibe) did not cause a lower risk of SLE.Conclusion: Evolocumab might provide a protective effect on the risk of SLE in the European population, but statins and ezetimibe might not have the protective effect. Further research is necessary to elucidate the specific mechanisms and potential therapeutic applications of PCSK9 inhibitors (evolocumab) in the context of SLE protection

    Utilization of antenatal ultrasound scan and implications for caesarean section: a cross-sectional study in rural Eastern China

    <p>Abstract</p> <p>Background</p> <p>Antenatal ultrasound scan is a widely accepted component of antenatal care. Studies have looked at the relationship between ultrasound scanning and caesarean section (CS) in certain groups of women in China. However, there are limited data on the utilization of antenatal ultrasound scanning in the general population, including its association with CS. The purpose of this study is to describe the utilization of antenatal ultrasound screening in rural Eastern China and to explore the association between antenatal ultrasound scan and uptake of CS.</p> <p>Methods</p> <p>Based on a cluster randomized sample, a total of 2326 women with childbirth participated in the study. A household survey was conducted to collect socio-economic information, obstetric history and utilization of maternal health services.</p> <p>Results</p> <p>Coverage of antenatal care was 96.8% (2251/2326). During antenatal care, 96.1% (2164/2251) women received ultrasound screening and the reported average number was 2.55. 46.8% women received at least 3 ultrasound scans and the maximum number reached 11. The CS rate was found to be 54.8% (1275/2326). After adjusting for socio-demographic and clinical variables, it showed a statistically significant association between antenatal ultrasound scans and uptake of CS by multivariate logistic regression model. High husband education level, high maternal age, having previous adverse pregnant outcome and pregnancy complications during the index pregnancy were also found to be risk factors of choosing a CS.</p> <p>Conclusions</p> <p>A high use of antenatal ultrasound scan in rural Eastern China is found and is influenced by socio-demographic and clinical factors. Evidence-based guidelines for antenatal ultrasound scans need to be developed and disseminated to clinicians including physicians, nurses and sonographers. Guidance about the appropriate use of ultrasound scans should also be shared with women in order to discourage unreasonable expectations and demands. It is important to monitor the use of antenatal ultrasound scan as well as the indications for caesarean section in rural China.</p

    MicroRNA-381 Negatively Regulates TLR4 Signaling in A549 Cells in Response to LPS Stimulation

    It is widely reported that miR-381 is dysregulated in various tumors. However, the specific role of miR-381 in respiratory infections has not been reported. To probe this role, A549 cells were pretreated with 1 μg/mL LPS for 24 h. The level of miR-381 was detected using RT-qPCR. The expression of proinflammatory cytokines was determined using an ELISA kit and western blotting. Bioinformatics analysis was used to predict the target genes of miR-381, and a luciferase reporter assay was used to validate the expression of the target genes. miR-381 expression was increased in A549 cells treated with LPS, which is a ligand of TLRs. Further study revealed that the overexpression of miR-381 increased the activity of NF-κB signaling, thereby increasing the expression of IL-6, TNFα, and COX-2. Further study revealed that IκBα was a target gene of miR-381. The upregulation of miR-381 under LPS stimulation contributes to respiratory infections mainly by targeting IκBα

    A meta-analysis of the efficacy and safety of adjuvant sorafenib for hepatocellular carcinoma after resection

    Abstract Background Sorafenib was reported as a useful adjuvant treatment in patients with hepatocellular carcinoma who underwent surgical resection. However, its therapeutic value remains controversial. This meta-analysis examined the available data regarding the efficacy and safety of sorafenib in patients with hepatocellular carcinoma after radical surgery. Methods The meta-analysis was conducted according to the Preferred Reporting Items for Systematic Reviews and Meta-Analyses guidelines. The protocol was registered in advance with PROSPERO (CRD42021233868). We searched PubMed, Embase, Cochrane Library, and Web of Science to identify eligible studies. Overall survival, recurrence-free survival, and recurrence rates were analyzed, and adverse events were reviewed. Hazard ratios or pooled risk ratios with 95% CIs were collected and analyzed using STATA version 12.0 in a fixed-effects or random-effects meta-analysis model. Results In total, 2655 patients from 13 studies were ultimately included in this meta-analysis. The combined results illustrated that sorafenib was associated with better overall survival than the control (hazard ratio = 0.71, 95% CI = 0.59–0.86; P < 0.001). Similarly, the drug also improved recurrence-free survival (hazard ratio = 0.68, 95% CI = 0.54–0.86, P = 0.001). Combined data revealed that patients treated with sorafenib after resection had a lower recurrence rate (pooled risk ratio = 0.78, 95% CI = 0.68–0.90, P < 0.001). The primary adverse events were hand-foot skin reaction, fatigue, and diarrhea of mild-to-moderate severity, whereas grade 4 adverse events were rare (< 1%). Conclusions This meta-analysis demonstrated that adjuvant sorafenib therapy after resection in patients with hepatocellular carcinoma could prolong overall survival and recurrence-free survival and reduce recurrence rates without intolerable side effects. However, more evidence is needed before reaching a definitive conclusion

    Novel transient-energy-based directional pilot protection method for HVDC line

    Abstract This paper proposes a novel directional pilot protection method based on the transient energy for bipolar HVDC line. Supposing the positive direction of current is from DC bus to the DC line, in the case of an internal line fault, the transient energies detected on both sides of the line are all negative within a short time, which denotes the positive direction fault; while for an external fault, the transient energy on one end is positive, which means a negative direction fault, but the transient energy on the opposite end is negative, which indicates that the fault direction is positive. According to these characteristics, an integration criterion identifying fault direction is constructed. In addition, through setting a fixed energy threshold, the faulted line and lightning disturbance can also be discriminated. Simulation results from UHVDC transmission system show the validity of the proposed protection method
